Digenic DUOX1 and DUOX2 Mutations in Cases With Congenital Hypothyroidism

The DUOX2 enzyme generates hydrogen peroxide (H2O2), a crucial electron acceptor for the thyroid peroxidase–catalyzed iodination and coupling reactions mediating thyroid hormone biosynthesis. DUOX2 mutations result in dyshormonogenetic congenital hypothyroidism (CH) that may be phenotypically heterogeneous, leading to the hypothesis that CH severity may be influenced by environmental factors (e.g., dietary iodine) and oligogenic modifiers (e.g., variants in the homologous reduced form of NAD phosphate-oxidase DUOX1). However, loss-of-function mutations in DUOX1 have not hitherto been described, and its role in thyroid biology remains undefined.

Tryptase-catalyzed core histone truncation: A novel epigenetic regulatory mechanism in mast cells

Mast cells are key effector cells in allergic reactions. When activated to degranulate, they release a plethora of bioactive compounds from their secretory granules, including mast cell-restricted proteases such as tryptase. In a previous study, we showed that tryptase, in addition to its intragranular location, can be found within the nuclei of mast cells where it truncates core histones at their N-terminal ends. Objective: Considering that the N-terminal portions of the core histones constitute sites for posttranslational modifications of major epigenetic impact, we evaluated whether histone truncation by tryptase could have an impact on epigenetic events in mast cells.

Molecular and cellular mechanisms linking inflammation to insulin resistance and b-cell dysfunction

Obesity is a major public health problem worldwide, and it is associated with an increased risk of developing type 2 diabetes. It is now commonly accepted that chronic inflammation associated with obesity induces insulin resistance and b-cell dysfunction in diabetic patients. Obesity-associated inflammation is characterized by increased abundance of macrophages and enhanced production of inflammatory cytokines in adipose tissue. Adipose tissue macrophages are suggested to be the major source of local and systemic inflammatory mediators such as tumor necrosis factor a, interleukin (IL)-1b, and IL-6. These cytokines induce insulin resistance in insulin target tissues by activating the suppressors of cytokine signaling proteins, several kinases such as c-Jun N-terminal kinase, IkB kinase b, and protein kinase C, inducible nitric oxide synthase, extracellular signal-regulated kinase, and protein tyrosine phosphatases such as protein tyrosine phosphatase 1B. These activated factors impair the insulin signaling at the insulin receptor and the insulin receptor substrates levels. The same process most likely occurs in the pancreas as it contains a pool of tissue-resident macrophages. High concentrations of glucose or palmitate via the chemokine production promote further immune cell migration and infiltration into the islets. These events ultimately induce inflammatory responses leading to the apoptosis of the pancreatic b cells. In this review, the cellular and molecular players that participate in the regulation of obesity-induced inflammation are discussed, with particular attention being placed on the roles of the molecular players linking inflammation to insulin resistance and b-cell dysfunction.

The effect of tamoxifen on endometrial carcinogenesis stems from its estrogen agonist effect.An in vivo study was carried out to compare the effect of tamoxifen-loaded solid lipid nanoparticles andfree drug on the ER- and VEGF-A genes expression.Material and methods: Twenty-four female Sprague-Dawley rats divided into 4 groups of six rats wereused for this study. The first and second groups were ovariectomized and given tamoxifen and tamoxifen-loaded SLN respectively for six days continuously. Group 3 served as the untreated ovariectomized controlgroup and group 4 was made up of untreated normal healthy rats. At the end of the study, the rats weresacrificed and study of the genes expression and serum oxidative stress were carried out.

Increased Red Blood Cell Volume Distribution Width: Important Clinical Implications in Predicting Gastric Diseases

The red blood cell distribution width (RDW) is a blood analyzer marker showing the peripheral blood erythrocyte volume heterogeneity parameters. It is a normal diagnosis index of many diseases. This study was performed to evaluate the relationship between the RDW and gastric diseases. Methods: A total of 189 patients with GC, 68 patients with gastric ulcers, 92 patients with chronic gastritis, and 157 healthy controls were enrolled in this study. Each patient’s RDW and other biomarkers were recorded. All of the statistical analyses and comparisons between each group were determined using SPSS16.0 software. The statistical significance level was set to a p-value < 0.05.

Tapeworm Diphyllobothrium dendriticum (Cestoda)— Neglected or Emerging Human Parasite?

A total number of 14 valid species of Diphyllobothrium tapeworms have been described in literature to be capable of causing diphyllobothriosis, with D. latum being the major causative agent of all human infections. However, recent data indicate that some of these infections, especially when diagnosed solely on the basis of morphology, have been identified with this causative agent incorrectly, confusing other Diphyllobothrium species with D. latum. Another widely distributed species, D. dendriticum, has never been considered as a frequent parasite of man, even though it is found commonly throughout arctic and subarctic regions parasitizing piscivorous birds and mammals. Recent cases of Europeans infected with this cestode called into question the actual geographic distribution of this tapeworm, largely ignored by medical parasitologists

: Malondialdehyde low-density lipoprotein (MDA-LDL) is a major form of oxidized LDL and considered to be more atherogenic than LDL. Information on major determinants of MDA-LDL and their association in subjects who are not under treatment for diabetes mellitus and dyslipidemia is limited. Methods: This study included 778 Japanese subjects who were not taking medication for diabetes mellitus and dyslipidemia. All subjects underwent an annual health examination that included MDA-LDL analysis. Study subjects were divided into four groups according to mean values of LDL-C and MDA-LDL, and the metabolic profile was compared.
